However, differential mutation rates in different partitions of the mitogenome may produce incongruent results. In this study, we sequenced 123 mitogenomes of 7 subspecies from lineages A, M, and C. This allowed generating a comprehensive dataset to investigate the phylogenetic and phylogeographic congruence among the mitogenome, individual genes, and the tRNAleu-COX2 region. We showed that the diversity patterns inferred from the tRNAleu-COX2 marker are not fully paralleled by those obtained with the mitogenome and the individual genes; while the three lineages are supported by these, the African sub-lineages and the haplotypes are not. Thus, conclusions drawn from the tRNAleu-COX2 region need to be taken with caution and this marker may not be appropriate to infer phylogenetic relationships between honey bee colonies. La región intergénica tRNAleu-COX2 del ADN mitocondrial se ha utilizado para evaluar la diversidad en poblaciones de abejas melíferas (Apis mellifera L.) de todo el mundo. Sin embargo, las tasas de mutación diferenciales en distintas particiones del mitogenoma pueden producir resultados incongruentes. En este estudio, secuenciamos 123 mitogenomas de 7 subespecies de los linajes A, M y C. Esto permitió generar un amplio conjunto de datos para investigar la congruencia filogenética y filogeográfica entre el mitogenoma, los genes individuales y la región tRNAleu-COX2. Demostramos que los patrones de diversidad inferidos a partir del marcador tRNAleu-COX2 no son totalmente paralelos a los obtenidos con el mitogenoma y los genes individuales; mientras que los tres linajes están apoyados por éstos, los sublinajes africanos y los haplotipos no lo están. Así pues, las conclusiones extraídas de la región tRNAleu-COX2 deben tomarse con cautela y este marcador puede no ser apropiado para inferir relaciones filogenéticas entre colonias de abejas melíferas. |
